The delta D temperature proxy in Antarctic ice cores varies in parallel with CO2 through glacial cycles. However, these variables display a puzzling asynchrony. Well-dated records of Southern Ocean temperature will provide crucial information because the Southern Ocean is likely key in regulating CO2 variations. Here, we perform multiple isotopic analyses on an Antarctic ice core and estimate temperature variations at this site and in the oceanic moisture source over the past 720,000 years, which extend the longest records by 300,000 years. Evaluation of novel oocyst wall protein candidates of Toxoplasma gondii OAK

Oocyst stage of Toxoplasma gondii is characterized by a durable wall that confers a strong protection to this protozoan parasite in face of harsh environmental conditions. Thus, it is considered the key for transmission of T. gondii. Analysis of oocyst wall composition is mandatory therefore; the aim of this study was to identify novel T. gondii oocyst wall proteins and test their use in detection of these oocysts in environmental samples. Five candidates of novel T. gondii oocyst wall proteins (TgOWPs) were identified and named TgOWP8 through TgOWP12. Neospora caninum infection is an important disease affecting bovine and canine populations worldwide. The dense granule antigen 7 of N. caninum (NcGRA7) is considered to be an immunodominant antigen. In the present study, the gene encoding truncated NcGRA7 (NcGRA7t) lacking an N-terminal signal peptide was expressed in Escherichia coli, and its diagnostic potential in an enzyme-linked immunosorbent assay (ELISA) was evaluated. The ELISA could clearly discriminate between known N. caninum-positive and -negative sera from dogs. In the asexual stages, Toxoplasma gondii stage converts between acute phase rapidly replicating tachyzoites and chronic phase slowly dividing bradyzoites. Correspondingly, T. gondii differentially expresses two distinct genes and isoforms of the lactate dehydrogenase enzyme, expressing LDH1 exclusively in the tachyzoite stage and LDH2 preferentially in the bradyzoite stage. Gastrointestinal zoonotic helminths of dogs and cats have a public health concern worldwide. We investigated the prevalence of gastrointestinal helminths of zoonotic significance in dogs and cats in lower Northern Thailand and utilized molecular tools for species identification of hookworms and Opisthorchis viverrini. Fecal samples of 197 dogs and 180 cats were collected. Overall prevalence of infection using microscopy was 40.1% in dogs and 33.9% in cats. The Influence of the regulation of Toxoplasma gondii TgMIC2 transgene on host cell infection OAK

Toxoplasma gondii microneme protein 2 (TgMIC2), an apically stored adhesin, was shown to be a key participant involved in the initial attachment and invasion to a host cell. In this study, we had established the clonal line of T. gondii tachyzoites which over-expressed TgMIC2 using the tetracycline repressor (TetR)-based inducible gene expression system. The TgMIC2 over-expression significantly reduced tachyzoite propagation in vitro (p < 0.002) and significantly reduced the parasite virulence in mice (p = 0.002). The over-expression of exogenous TgMIC2 caused the increase of mRNA expression level of endogenous TgMIC2. A combination of antigenic regions of microneme proteins have been previously reported as being protective against chronic toxoplasmosis. In this work, we evaluated immune responses induced by immunizing BALB/c and C57BL/6 mice intradermally with plasmid DNA encoding the protein sequences of Toxoplasma gondii AMA1, MIC2, M2AP and BAG1. Mice immunized with the AMA1 gene developed high levels of serum IgG2a and c antibodies as well as cellular immune responses associated with IFN-γ synthesis suggesting a modulated Th1 type of response. Immunization with the AMA1 gene resulted in a partial but significant protection against the acute phase of toxoplasmosis compared to MIC2, M2AP and BAG1 genes.
